Gene Expression Profiles

Using our qRT-PCR platform we provide robust, quantitative gene expression profiles in standard and custom cohorts of diseased and non-diseased human tissues. We are also able to characterize tissue cohorts for specific somatic mutations, like KRAS, EGFR, AKT using a qRT-PCR based approach.

XpressWay™ Gene Expression Profiles

Asterand Bio’s XpressWay gene expression profiles are quantitative gene transcripts expression patterns across 72 tissues (3 individual donors of each) representing the major organ systems of the human body, providing a reproducible and reliable rigid assay that is essential for target validation, safety profiling and validation of animal models.

Using a rigorously controlled process, from tissue to data, Asterand Bio has used the same highly sensitive, robust quantitative real-time PCR (qRT-PCR) methodology for the last 20 years ensuring reproducibility and reliability. To ensure reliability and repeatability across assays, we utilize a global standard curve inclusive of 81 genes, 150 individual standard curves and 1476 data points.

Stellar plots provide a visual representation of the expression levels (mean copy number) for all of the 72 tissues. Each “spoke of the wheel” represents one tissue type, grouped by biological system, with the expression level being lowest in the middle and highest on the outside. The software autoscales the outer perimeter based on the highest copy number (Cn) expressed for a given gene profile.

XpressWay Profile Reports

Data are provided in the format of PDF reports containing the relevant information regarding the target gene, data and donor details. The raw data for the target gene and GAPDH, together with the donor details, are also provided in Excel format to enable the data to be presented in any format required by the client.

Also included are correlation coefficients calculated by comparing the expression profile of the target gene with the profiles of all the other genes in Asterand’s XpressWay profile database.

Details for the construction of the global standard curve, used to convert cycle thresholds (Ct values) to copy number, are also provided.

Example XpressWay Profiles

XpressWay profiles can be used to illustrate the expression of specific mRNAs. As can be seen below, expression patterns can clearly delineate cardiac vs. CNS vs. gastrointestinal expression as well as restricted expression (non-CNS).

XpressWay Profiles – Splice Variant Expression

XpressWay profiles illustrate that different profiles can be obtained for gene splice variants, demonstrating differences in distribution.

One of the 5HT receptors, profiled using a generic primer–probe set (top left) picks up multiple splice variants and demonstrates expression is predominantly Gastrointestinal and CNS.

To further delineate specific expression of splice variants, different primer-probe sets that are specific for 4 different splice variants of this 5HT receptor were designed and utilized to demonstrate different expression patterns:

  • SV2 is very similar to the generic profile
  • SV1 generally shows broader pattern of expression and is high in CV tissues (potential safety liability if expression is functional)
  • SV3 is very GI restricted expression
  • SV4 is very CNS restricted expression
